丁酰胆碱酯酶(Butyrylcholinesterase,BchE,EC3.1.1.8),又称血浆胆碱酯酶,假性胆碱酯酶,是一种丝氨酸水解酶,由肝脏合成后进入血液,几乎存在于所有动物组织中。BchE结构与乙酰胆碱酯酶(AchE)相似,但底物特异性和抑制剂敏感性不同。与AchE相比,BchE能够有效水解较大的胆碱酯,如丁酰胆碱和苯甲酰胆碱,而且可以清除有机磷类农药、氨基甲酸酯类农药等神经毒剂的毒害作用。有研究表明,BchE可作为阿尔茨海默病治疗的重要靶点。BchE催化丁酰胆碱水解生成胆碱,胆碱与二硫对硝基苯甲酸(DTNB)作用生成5-巯基-硝基苯甲酸(TNB);TNB在412nm处有吸收峰,通过测定412nm吸光度增加速率,计算BchE活性。

注意:实验之前建议选择2-3个预期差异大的样本做预实验。如果样本吸光值不在测量范围内建议稀释或者增加样本量进行检测。
需自备的仪器和用品:
可见分光光度计、低温离心机、分析天平、水浴锅/恒温培养箱、1mL玻璃比色皿、可调式移液枪、研钵/匀浆器/细胞超声破碎仪、冰和蒸馏水。
操作步骤:
一、样本处理(可适当调整待测样本量)
1. 组织样本:按照组织质量(g):试剂一体积(mL)=1:5~10比例加入试剂一(建议称取0.1g样本,加入1.0mL试剂一),冰浴匀浆后,于4℃,12000rpm离心10min,弃沉淀,取上清液置于冰上待测。
2. 血清/血浆等液体样本:直接测定。若有浑浊请离心后取上清置于冰上待测。
3. 细菌、细胞:按照细胞数量104个:试剂一体积(mL)500~1000:1的比例(建议500万细胞加入1mL试剂一),冰浴超声波破碎细胞(功率300w,超声3s,间隔7s,总时间3min),于4℃,12000rpm离心10min,弃沉淀,取上清液置于冰上待测。
