溶液的配制:
1、显色液:临用前根据样本数量按照试剂一:试剂二:试剂三=250μL:250μL:250μL(750μL,1T)的比例配制显色液,充分混匀,现配现用;
2、标准品:临用前加入0.71mL蒸馏水和10μL浓硫酸,配制成50μmol/mLFe2+标准液。溶解好的标准品2-8℃可以保存2周。
3、62.5nmol/mL标准溶液的配制:临用前取50μL50μmol/mLFe2+标准液和950μL蒸馏水混合配制成2.5μmol/mL标准溶液;再吸取25μL2.5μmol/mL(2500nmol/mL)和975μL蒸馏水混合配制成62.5nmol/mL标准溶液备用。
产品说明:
双子叶植物和非禾本科单子叶植物采用铁螯合还原反应的高效活化和吸收机制从土壤中获取铁。三价铁还原为二价铁后才能被植物吸收利用。在铁充足时,植物根部的三价铁氧化还原酶将Fe(III)-螯合物中的铁还原并将还原所得Fe2+转运通过细胞质膜进入根细胞内。高铁螯合物还原酶(Ferricchelatereductase,FCR,EC1.16.1.7)催化Fe3+还原为Fe2+,Fe2+和菲洛嗪形成紫色络合物,在562nm下有特征吸收峰。

注意:实验之前建议选择2-3个预期差异大的样本做预实验。如果样本吸光值不在测量范围内建议稀释或者增加样本量进行检测。
需自备的仪器和用品:
可见分光光度计、低温离心机、分析天平、1mL玻璃比色皿、可调式移液枪、研钵/匀浆器、浓硫酸(95%-99%AR)、冰和蒸馏水。
操作步骤 :
一、样本处理(可适当调整待测样本量)
1.组织样本:按质量(g):提取液体积(mL)1:5~10比例加入提取液(建议称取0.1g样本,加入1.0mL提取液),冰浴匀浆后,于4℃,8000g,离心10min,弃沉淀,取上清液置于冰上待测。
2.液体样本:直接测定。若液体有浑浊则离心取上清测定。
